Measles serodiagnosis: production and evaluation of the IgM-measles ELISA IAL reagent BJM

Recent measles outbreaks in different countries led to an increase of laboratory measles diagnosis. Thus, we developed the IgM-Measles ELISA IAL, using measles virus antigens obtained from cell cultured in microcarriers in order to supply reagent kits to Brazilian public health laboratories. A batch of antigenic reagent was produced and evaluated in the enzyme immunoassay in comparison with clinical diagnosis and with as reference assay (IgM Capture ELISA CDC) data. This study was performed in a positive panel with 70 serum samples from patients with measles, and a negative panel with 132 samples from patients with unrelated diseases and without recent measles or vaccination history. Performance evaluation of CHO-K1 cell in culture medium supplemented with hemolymph BABT

The aim of this work was to evaluate the potential of hemolymph utilization as a culture medium supplement to cultivate the animal cell CHO-K1. For this purpose 1% v/v of hemolymph was added to DMEM medium containing 10% v/v of FBS and 1 or 4.5 g/L of glucose. The culture was grown in spinner flasks incubated in a 10% v/v CO2 environment, at 37ºC, with the Cytodex 1 microcarrier. Comparing the results obtained from the culture with hemolymph against those without hemolymph, a positive influence of the hemolymph was observed, as the experiment with hemolymph presented a 52% higher cell concentration and a higher productivity of up to 40%.

The number of biopharmaceuticals for medical and veterinarian use produced in mammalian cells is increasing year after year. All of them are obtained by stable recombinant cell lines. However, it is recognized that transient gene expression produces high level expression in a short time. In that sense, viral vectors have been extensively used for producing recombinant proteins on lab-scale. Among them, Semliki Forest virus is commonly employed for this purpose. This review discusses the main aspects related to the use of Semliki Forest virus technology as well as its advantages and drawbacks which limit currently its utilization in biopharmaceutical industry on large-scale.
